I already have the twist-lok lines and fittings for my fuel system, I just wasn't sure if it was able to be used for transmission cooling as well.

When assembling my fuel system, I had no problem putting the line on the fittings, they are double barbed and you have to push like crazy to install. I have no worries about these coming lose, plus it was real quick to put together.
Ah, looks like that line is rated to 250psi and is oil safe, which should be plenty for trans cooler lines, and I am sure the ends would be fine too - the cooler lines on my Toyota (with similar pressures as a GM OD Trans) uses fuel injection rated clamps at the cooler end and oil-safe fuel injection hose... been great for 2+ years and 50,000 miles.
